Spelunky! is the latest pixel indie game darling from Derek Yu (Aquaria). Here's his descriptive paragraph:
"Probably the easiest way to describe Spelunky is that its (kind of) like La Mulana meets Nethack - every time you play the levels, items, monsters, and so forth, are all procedurally-generated. And the terrain is destructible and there are quite a few ways in which the various game elements can interact with one another."

I used to think Gamemaker wasn't that great of a developing platform, but after seeing this I'm amazed.

Never took game maker that seriously,
the best game I played made by it was made with the full version of game maker. |
That's a fun game - hard as heck, but fun. I love the procedurally generated part, too.
It does make me wonder, if someone came up with a decent platformer system for BYOND, would BYOND be able to handle a game like this? |

Single-player (as it is), probably yes, though I'm not sure if it would be just as smooth (even though you can freely decrease world.tick_lag to improve that).

Yes and no. BYOND could handle Spelunky, but unfortunately it couldn't handle the simultaneous keys that you tend to need in Spelunky. BYOND's almost to the point where it can make a really ripping single-player action game, but not quite there yet.

BYOND's key press/release macro stuff can't handle multiple keys pressed at once?

It can. The only small problem with BYOND's macros is that they're a little slow, but Kakashi24124's K_KeyHandling library fixes that (might have gotten his key wrong).

I believe Game Maker is quite similar to BYOND, I tend to use it from time to time and I have yet to find any major limitations. It seems like most of their developers are a little more serious about developing games than our members.
